  • 简介:BACKGROUND:Alongwithaging,antioxidaseactivitydecreasesandoxygen-derivedfreeradicalsgreatlyaccumulate,resultingincellularsenescence,orevencelldeath.Thisismanifestedbyhypomnesiaanddis-orderedmetabolismoffreeradicals.StudieshavereportedthatLongyanshenpolysaccharideshavethefunc-tionofantioxidationandimprovedbrainmemory.OBJECTIVE:ToobservetheeffectsofLongyanshenpolysaccharidesonfreeradicalmetabolisminbraintissuetoverifytheanti-agingmechanismsinsenescenceaccelerated-prone(SAMP8)mice.DESIGN,TIMEANDSETTING:Therandomized,controlled,biochemicalexperimentwasperformedintheDepartmentofPharmacologyandScientificExperimentalCenterofGuangxiMedicalUniversity(China)fromSeptember2005toJanuary2008.MATERIALS:FortySAMP8micewererandomizedintofourgroups:SAMP8controlgroup,aswellaslow-,mid-,andhigh-dosepolysaccharide,with10miceineachgroup.Tensenescenceaccelerated-resistant-prone(SAMR1)miceservedasthenormalcontrolgroup.Longyanshenpolysaccharides,extractedfromthemedicalplantLongyanshen,weresuppliedbytheDepartmentofPharmacology,GuangxiMedicalUniversity.Superoxidedismutase(SOD),glutathioneperoxidase(GSH-Px),malonaldehyde(MDA),nitricoxide(NO),andtotalproteintestkitwerepurchasedfromNanjingJianchengBioengineeringInstitute(China).METHODS:SAMP8micewereusedtoestablishadementiaanimalmodel.SAMP8andSAMR1controlmicewereadministered30mL/kgsaline.Thelow-,middle-,andhigh-dosepolysaccharidegroupswereadministered45,90,and180mg/kgLongyanshenpolysaccharides,respectively.Eachgroupwastreatedbyin-tragastricadministration,oncedaily,for50continuousdays.MAINOUTCOMEMEASURES:Onehourafterthelastadministration,mousebraintissueswerecollected,andretroorbitalbloodsamplingwasperformed.SpectrophotometrywasusedtomeasureSODandGSH-Pxactivity,aswellasMDAandNOconcentrationinseraandbrainsofSAMP8mice.RESULTS:

  • 简介:Objective:TostudyexplorestheeffectofHLEConthesecretedproteinsofepithelialovariancancer(EOC)cells(SKOV3-PM4)withdirectionalhighlylymphaticmetastasis.Methods:Supernatantsoffourgroupsofculturedcells,namely,SKOV3(A),SKOV3+HLEC(B),SKOV3-PM4(C),SKOV3-PM4+HLEC(D),werecollected,andtheirproteinsweredetectedbyantibodyarraysandiTRAQ-2D-LC-MALDITOF/TOF/MS.SignificantlydifferentialproteinswerefurtheranalyzedviabioinformaticsandvalidatedinhumanserumsandcellmediaviaELISA.Results:ResultsofantibodyarraysandmassspectrometrydemonstratedthatGRNandVEGFAwereupregulatedingroupC(comparedwithgroupA),whereasIGFBP7andSPARCweredownregulatedingroupD(comparedwithgroupC).ComprehensivebioinformaticsanalysisresultsshowedthatIGFBP7andVEGFAwerecloselylinkedtoeachother.FurthervalidationwithserumsshowedstatisticalsignificanceinVEGFAandIGFBP7levelsamonggroupsofpatientswithovariancancers,benigntumors,andcontrolgroups.Twoproteinswereupegulatedinthefirstgroup.VEGFAinthecontrolgroupwasdownregulated.ForIGFBP,upregulationinthecontrolgroupanddown-regulationinthefirstgroupwerealsoobserved.Conclusion:TheHLECmicroenvironmentiscloselyassociatedwithdirectionalmetastasistolymphnodesandwithdifferentialproteinsincludingcellstromalproteinsandadhesionfactors.TheupregulationofVEGFAandGRNandthedownregulationofSPARCandIGFBP7arecloselyassociatedwithdirectionalmetastasistolymphnodesinEOCcells.

  • 简介:Toinvestigatethetreatmenteffectof2-seleniumbridgedβ-cyclodextrin(2-SeCD),aGPXmimic,onthestrokeofstroke-pronespontaneouslyhypertensiverats(SHRSP),fifty-twoSHRSPof8-weekoldwererandomlydividedintofourgroupsA,B,CandcontrolgroupD.TheratsofgroupsA,B,CandDweregiven1.0%-1.5%NaClmassfractionasdrinkingfluid.Afteronsetofstroke,groupsA,BandCweregivenorally16.05,160.5and1605mg*kg-1*day-1of2-SeCD,respectively,andgroupDwasgivenwaterfor2weeks.Theclinicalscoreofstroke,systolicbloodpressure(SBP),survivaltimeofratswererecordedandthehistopathologicexaminationsoftheirbrainandcarotidarteryweremadeafterdecapitation.Theclinicalscoresofstrokeaftertreatmentwith160.5mg*kg-1*day-1(GroupB)and1605mg*kg-1*day-1(GroupC)of2-SeCDare2.55±0.98and1.98±0.79,respectively,thoseareobviouslylowerthanthatofgroupD(3.41±0.83,p<0.01).ThesurvivaldaysingroupB(10.0±8.6)andgroupC(14.4±7.9)arelongerthanthatforgroupD(4.7±2.9,p<0.01).TheelectronmicroscopestudyshowedthattheendotheliumofcarotidarterywasneartonormalingroupBandgroupC,whileitwasseriouslyinjuredincontrolgroupDandmildlyinjuredingroupA.2-SeCDmayeffectivelybeusedtotreatthestrokeforSHRSP.

  • 简介:Basementmembranedegradationandblood-brainbarrierdamageappearaftercerebralinfarction,severelyimpactingneuronalandbrainfunctioning;however,theunderlyingpathogeneticmechanismsremainpoorlyunderstood.Inthisstudy,weinducedcerebralinfarctioninstrokepronespontaneouslyhypertensiveratsbyintragastricadministrationofhigh-sodiumwater(1.3%NaCl)for7consecutiveweeks.Immunohistochemicalandimmunofluorescenceassaysdemonstratedthat,comparedwiththenon-infarctedcontralateralhemisphere,stroke-pronespontaneouslyhypertensiveratsonnormalsodiumintakeandWistar-Kyotorats,matrixmetalloproteinase-9expression,thenumberofbloodvesselswithdiscontinuouscollagenIVexpressionandmicrovesseldensityweresignificantlyhigher,andthenumberofcontinuouscollagenIV-positivebloodvesselswaslowerintheinfarctborderzonesofstroke-pronespontaneouslyhypertensiveratsgivenhigh-sodiumwater.Linearcorrelationanalysisshowedmatrixmetalloproteinase-9expressionwaspositivelycorrelatedwiththenumberofdiscontinuouslycollagenIV-labeledbloodvesselsandmicrovesseldensityincerebralinfarctsofstroke-pronespontaneouslyhypertensiverats.Theseresultssuggestthatmatrixmetalloproteinase-9upregulationisassociatedwithincreasedregionalangiogenesisanddegradationofcollagenIV,themajorcomponentofthebasallamina,instroke-pronespontaneouslyhypertensiveratswithhigh-sodiumwater-inducedfocalcerebralinfarction.

  • 简介:Inthepresentstudy,theeffectofblockageofthecostimulatorysignalCD86attimeofimplantationontheexpressionsofTGF-β1,MMP-9,TIMP-3andPAI-1proteinsatthematernal-fetalinterfaceandtheoutcomeofpregnancyinmurineabortion-pronemodelwasinvestigated,inwhichtheCBA/JxDBA/2matingswereusedastheabortion-pronemodelandtheCBA/J×BALB/cmatingsusedasthenormalpregnantmodel.Thestudywasperformedinfollowingthreegroups:2groupsoftheabortion-pronemodel,whichwereexperimentalgroupandcontrolexperimentalgroup,and1groupofnormalpregnantmodel,andeachgrouphad10pregnantCBA/Jmiceexclusively.FemalepregnantCBA/Jmiceintheexperimentalgroupreceivedanintraperitoneal(i.p.)injectionof100μgofantimouseCD86mAbin200μlofPBSatday4.5ofgestation,andtheirrelevant-isotopematchedratIgG2bwasadministratedinthecontrolexperimentalgroupwiththesamedosageandatsametime.Forthenormalpregnantgroup,notreatmentwasgiven.ThepregnantCBA/Jmicewerekilledonday13.5ofgestation.Then,theembryoresorptionratewascalculatedandtheexpressionsofTGF-β1,MMP-9,TIMP3andPAI-1weredetectedbyusingimmunohistochemicalmethods.Itwasdemonstratedthattheembryoresorptionrateintheexperimentalgroupwassignificantlyreducedincomparisonwiththatinthecontrolexperimentalgroup(x2=7.441,P=0.006),buttherewasnosignificantdifferencewiththatinnormalpregnantgroup(x2=0.016,P=0.898).TheexpressionsofTGF-β1andPAI-1intheexperimentalgroupweresignificantlyincreasedincomparisonwiththatinthecontrolexperimentalgroup(P=0.010,P=0.003,respectively),withnosignificantdifferencefromthatinthenormalpregnantgroup(P=0.500).However,theexpressionofMMP-9intheexperimentalgroupwassignificantlyreducedincomparisonwiththatinthecontrolexperimentalgroup(P=0.012)withnosignificantdifferencefromthatinthenormalpregnantgroup(P=0.500).Theexpression
