简介:摘要通过1-methyl-3-nitro-1-nitroso-guanidin与coldplasma复合诱变Bacillussp.(兼性厌氧产丁醇芽孢杆菌)C2菌株,再利用分离筛选获得总溶剂产量与三株丁醇都有较为显著提高的突变菌株,突变菌株发酵10%木薯醪液之后总溶剂产量与丁醇分别达到20.79g/L、20.14g/L、20.36g/L与12.74g/L、12.44g/L、12.59g/L,与出发菌株相比较分别提高了15.4%-19.1%、21.6%-24.5%。优化编号414的突变菌株的发酵条件,在100mL的三角瓶发酵体系里面,编号为414的突变菌株的最佳发酵条件为接种量10%,种龄为24h,ph值控制在7左右,装液量为100mL,然后在37℃温度环境下静置发酵72h,在这个发酵条件之下,此菌株发酵7%木薯醪液产总溶剂量与丁醇分别达到了26-30g/L,17-19g/L。
