Aflexibleapproachtoethyl(3R,4S)-N-Boc-4-amino-3-hydroxy-5-phenylpentanoate(N-Boc-AHPPA-OEt),theγ-amino-β-hydroxyacidmoietyofhapalosinisdescribed.Thesyntheticmethodfeaturesaring-openingethanolysisofanactivatedN-Boc-lactam,whichisobtainedviaadiastereoselectivereductive-alkylationof(R)-malimidederivative.TheflexibilityofthemethodresidesintheintroductionofthealkylsidechainbyGrignardreagentaddition.
