  • 简介:Objective:Toevaluatetheeffectofosteoporosisonfracturehealingthroughobservingthehistomorphologicalchanges,bonemineraldensityofcallusandexpressionanddistributionoftransforminggrowthfactorbeta1(TGF-β1),basicfibroblastgrowthfactor(bFGF)andbonemorphogeneticprotein-2(BMP-2)inovariectomizedrats.Methods:SixtyfemaleSprague-Dawleyrats(aged12weeksandweighing235gonaverage)wererandomlydividedintoanovariectomized(OVX)group(n=30)andasham-operated(SO)group(n=30).OvariectomywasperformedintheOVXratsandsameincisionwasmadeintheSOrats.Threemonthslater,fractureoffemoralshaftwasmadeonalltherats.Thentheywerekilledatdifferenttimepoints.Callusformationwasobservedwithhistologicalandimmunohistochemicalmethods.Results:AreductionincallusandbonemineraldensityinthehealingfemurandadecreaseofosteoblastsexpressingTGF-β1nearthebonetrabeculawereobservedintheOVXrats3-4weeksafterfracture.HistomorphologicalanalysisrevealedahighercontentofsoftcallusintheOVXratsthanthatintheSOrats.ImmunohistochemistryresultsshowedthatnoremarkabledifferenceinexpressionanddistributionofBMP-2andbFGFbetweentheOVXandSOgroupswasfound.Conclusions:Osteoporosisinfluencesthequantityandqualityofcallusduringtheearlyperiodoffracturehealing.TheeffectofosteoporosisonfracturehealinghasnorelationshipwiththeexpressionofBMP-2orbFGF.ThedecreasedexpressionofTGF-β1inosteoblastsmaycauseadecreaseinqualityoffacturehealingafterosteoporosis.

  • 简介:AIM:Toinvestigatewhetherthesimultaneoustreatmentwithhumangrowthhormone(hGH)abolishesthenegativeeffectsofeverolimusonanastomotichealing.METHODS:Forty-eightmaleSprague-Dawley-ratswererandomizedtothreegroupsof16animalseach(Ⅰ:vehicle;Ⅱ:everolimus3mg/kgpo;Ⅲ:everolimus3mg/kgpo+hGH2.5mg/kgsc).AnimalswerepretreatedwithhGHand/oreverolimusdailyforsevendays.Thenastandardanastomosiswascreatedinthedescendingcolonandtreatmentwascontinuedforanothersevendays.Theanastomosiswasresectedintotoandtheburstingpressurewasassessedasamechanicalparameterofintestinalhealing.Moreover,biochemical(Hydroxyproline,PCNA,MPO,MMP-2andMMP-9)andhistological(celldensity,angiogenesis,amountofgranulationtissue)parametersofintestinalhealingwereassessed.RESULTS:AnastomoticburstingpressurewassignificantlyreducedbyeverolimusandasimultaneoustreatmentwithhGHresultedinconsiderablyhighervalues(Ⅰ:134±19mmHg,Ⅱ:85±25mmHg,Ⅲ:114±25mmHg;P<0.05,ⅠvsⅡ;P=0.09,ⅠvsⅢandⅡvsⅢ)HydroxyprolineconcentrationwassignificantlyincreasedbyhGHcomparedtoeverolimusalone(Ⅰ:14.9±2.5μg/mg,Ⅱ:8.9±3.6μg/mg,Ⅲ:11.9±2.8μg/mg;P<0.05,?ⅠvsⅡ/ⅢandⅡvsⅢ).ThenumberofMPO-positivecellswasreducedsignificantlybyhGHcomparedtoeverolimusalone(Ⅰ:10±1n/mm~2,Ⅱ:15±3n/mm~2,Ⅲ:9±2n/mm~2;P<0.05,ⅠvsⅡandⅡvsⅢ),whilethenumberofPCNA-positivecellswereincreasedbyhGH(Ⅰ:28±3/mm~2,Ⅱ:12±3/mm~2,Ⅲ:26±12/mm~2;P<0.05,?Ⅰ?vsⅡandⅡvsⅢ).Correspondingtothesebiochemicalfindings,HEhistologyrevealedsignificantlyincreasedamountofgranulationtissueinhGH-treatedanimals.CONCLUSION:InhibitionofintestinalwoundhealingbyeverolimusispartiallyneutralizedbysimultaeoustreatmentwithhGH.BothinflammationaswellascollagendepositionisinfluencedbyhGH.

  • 简介:Toevaluatetheinfluenceofosteoporosisonthemiddleandlateperiodsoffracturehealingprocessthroughobservingthehistomorphologicalchanges,honemineraldensityandbiomechanicalpropertiesinovariectomizedrats.Methods:Eighty-fourfemaleSDratsof4monthsoldwererandomlydividedintoosteoporosisgroupandshamoperationgroup,42ineach.Ratsinosteoporosisgroupwereperformedovariectomyoperationwhilethoseinshamoperationgroupweregivenshamoperation.Amidshafttibiafracturemodelwasestablished10weeksafterovariectomy.Tibiaswereharvested2,4,6,12,18weeksafterfractureforbonemineraldensity,histomorphologicalandbiomechanicalevaluation.Results:Comparedwiththeshamoperationgroup,callusbonemineraldensitywas12.8%,18.0%,17.0%lowerinosteoporosisgroup6,12,18weeksafterfracture,respectively(P<0.05);callusfailureloadwas24.3%,31.5%,26.6%,28.8%lowerinosteoporosisgroup,andcallusfailurestresswas23.9%,33.6%,19.1%,24.9%lowerinosteoporosisgroup4,6,12,18weeksafterfracture,respectively(P<0.05).Inosteoporosisgroup,endochondralboneformationwasdelayed,moreosteoclastcellscouldbeseenaroundthetrabecula,andthenewbonetrabeculaarrangedlooselyandirregularly.Conclusions:Osteoporosisinfluencesthemiddleandlateperiodsoffracturehealingintheratosteoporoticmodel.Theimpairmentisconsideredtobetheresultofcombinedeffectsofprolongedendochondralcalcification,highactivatedosteoclastcellandthedecelerationoftheincreaseinbonemineraldensity.

  • 简介:Objective:Tostudythechangeofthestressshieldingrateofstress-relaxationplateinvivoanditsinfluenceonfracturehealing.Methods:Thediaphysesofbilateraltibiasin70NewZealandrabbitswereosteotomizedandfixedwithstressrelaxationplates(SRP,theSRPgroup)andrigidplates(RP,theRPgroup),respectively.Thefracturehealingprocessinthese2groupswasinvestigatedbyradiography,lightandpolarizedlightmicroscopyandbiomechanicaltestat2to48weekspostoperatively.Results:Earlyafterfixationthestressshieldingratewasabout70%inbothgroups.WhileintheSRPgroupthestressshieldingratedecreasedgraduallyastimepassed,whyichwassignificantlylowerthanthatoftheRPgroup(P<0.05)bytheendofthe8thpostoperativeweek,andstabilizedatthelevelofabout27%at36-48weeksafterfixation.AbundantexternalcallusassociatedwiththeformationofcartilaginouscalluscouldbeobservedintheSRPgroupat2-4weekspostoperatively.Thetransformationofthecallusintothelamellarbonebeganat8-12weeks,thecollagengraduallyarrangedinorder,andthemechanicalnatureoftheunitedbonewasgraduallystrengthened,too.IntheRPgroup,theexternalcalluswasscarceattheearlystageoffracturehealing,andthecallusremodelingatthelatestageoffracturehealingwasdominatedbyboneabsorption.Theultimatebendingstrength(UBS)wasonly57.95%ofthatofthenormalby48weeks.Conclusions:ThedecreaseofthestressshieldingrateofSRPinvivowaswellinterrelatedwiththetimeoffixation.TheapplicationofSRPcouldpromotethecallusformationandbonereconstructionthustofavortherecoveryofthemechanicalstrengthoftheunitedbone.
