Isolation and characterization of the murine Nanog gene promoter-中国期刊网

摘要 Nanogproteinisexpressedintheinteriorcellsofcompactedmorulaeandmaintainedtillepiblastsbutdownregulatedbyimplantationstage.Itisalsoexpressedinembryonicstemcells,embryoniccarcinomacellsandembryonicgermcellsbutdisappearedindifferentiatedEScells.Inthisstudy,wehaveisolated,sequenced,andperformedthefirstcharacterizationoftheNanogpromoter.Thetranscriptionstartsitesweremappedbyprimerextensionanalysis.TwopromoterregionswerefoundupstreamthetranscriptionstartsitesandtheexpressionofmajorNanogpromoter/reportergeneconstructisabolishedindifferentiatedF9ECcellsascomparedtotheundifferentiatedcounterpart.Wealsoshowedthataputativeoctamermotif(ATGCAAAA)isnecessaryforthemajorpromoteractivity.GelshiftandsupershiftassaysshowedthatOct-1,Oct-4andOct-6proteinselectivelybindtotheoctamermotif.
