Microsatellite-Centromere Mapping in Japanese Scallop(Patinopecten yessoensis) Through Half-Tetrad Analysis in Gynogenetic Diploid Families

摘要 Gene-centromeremappingisanessentialprerequisiteforunderstandingthecompositionandstructureofgenomes.Half-tetradanalysisisapowerfultoolformappinggenesandunderstandingchromosomalbehaviorduringmeiosis.TheJapanesescallop(Patinopectenyessoensis),acold-tolerantspeciesinhabitingthenorthwesternPacificcoast,isacommerciallyimportantmarinebivalveinAsiancountries.Inthisstudy,inheritanceof32informativemicrosatellitelociwasexaminedin70-hD-shapedlarvaeofthreeinducedmeiogynogeneticdiploidfamiliesofP.yessoensisforcentromeremappingusinghalf-tetradanalysis.Theratioofgynogeneticdiploidswasproventobe100%,100%and96%inthethreefamilies,respectively.Inheritanceanalysisinthecontrolcrossesshowedthat51ofthe53genotypicratiosobservedwereinaccordancewithMendelianexpectationsatthe5%levelafterBonferronicorrection.Sevenofthe32microsatellitelocishowedtheexistenceofnullallelesincontrolcrosses.Theseconddivisionsegregationfrequency(y)ofthemicrosatellitelocirangedfrom0.07to0.85withameanof0.38,suggestingtheexistenceofpositiveinterferenceafterasinglechiasmaformationinsomechromosomesinthescallop.Microsatellite-centromeredistancesrangedfrom4cMto42cMundertheassumptionofcompleteinterference.Informationonthepositionsofcentromeresinrelationtothemicrosatellitelociwillrepresentacontributiontowardstheassemblyofgeneticmapsinthecommerciallyimportantscallopspecies.
