Percoll fractionation of adult mouse spermatogonia improves germ cell transplantation

Aim:Toisolateandtransplantgermcellsfromadultmousetestesfortransplantation.Methods:Inordertodistinguishtransplantedcellsfromendogenouscellsofrecipients,donortransgenicmiceexpressinggreenfluorescentprotein(GFP)wereused.Germcellswerecollectedfromthedonorsat10-12weeksofageandspermatogoniawereconcentratedbypercollfractionationandtransplantedintorecipientseminiferoustubulesthathadbeenpreviouslytreatedwithbusulfanat5weeksofagetoremovetheendogenousspermatogeniccells.Results:Twentyweeksafterthetransplantation,awidespreadGFPsignalwasobservedintherecipientseminiferoustubules.Thepresenceofspermatogenesisandspermatozoawasconfirmedinsectionsof12outof14testestransplanted(86%).However,whengermcellsweretransplantedwithoutconcentrationthesuccessratewaszero(0/9).Conclusion:Germcellsfromadultmousetestescanbesuccessfullytransplantedintorecipientseminiferoustubulesifthecellpopulationisrichinspermatogoniaandthepercollfractionationisusefulinobtainingsuchacellpopulation.
