  • 简介:AbstractBackground:Immune- and inflammation-related genes (IIRGs) play an important role in the pathogenesis of tuberculosis (TB). However, the relationship between IIRG polymorphisms and TB risk remains unknown. In this study, the gene polymorphisms and their association with tuberculosis were determined in a Chinese population.Methods:We performed a case-control study involving 1016 patients with TB and 507 healthy controls of Han Chinese origin. Sixty-four single-nucleotide polymorphisms (SNPs) belonging to 18 IIRGs were genotyped by the PCR-MassArray assay, and the obtained data was analyzed with χ2-test, Bonferroni correction, and unconditional logistic regression analysis.Results:We observed significant differences in the allele frequency of LTA rs2229094*C (P = 0.015), MBL2 rs2099902*C (P = 0.001), MBL2 rs930507*G (P = 0.004), MBL2 rs10824793*G (P = 0.004), and IL12RB1 rs2305740*G (P = 0.040) between the TB and healthy groups. Increased TB risk was identified in the rs930507 G/G genotype (Padjusted = 0.027) under a codominant genetic model as well as in the rs2099902 (C/T + C/C) vs T/T genotype (Padjusted = 0.020), rs930507 (C/G + G/G) vs C/C genotype (Padjusted = 0.027), and rs10824793 (G/A + G/G) vs A/A genotype (Padjusted = 0.017) under a dominant genetic model after Bonferroni correction in the analysis of the overall TB group rather than the TB subgroups. Furthermore, the rs10824793_rs7916582*GT and rs10824793_rs7916582*GC haplotypes were significantly associated with increased TB risk (P = 0.001, odds ratio [OR] = 1.421, 95% confidence interval [CI]: 1.152-1.753; and P = 0.018, OR = 1.364, 95% CI: 1.055-1.765, respectively). Moreover, the rs10824793_rs7916582*AT/AT or rs10824793_rs7916582*GT/GT diplotype showed a protective (P = 0.003, OR = 0.530, 95% CI: 0.349-0.805) or harmful (P = 0.009, OR = 1.396, 95% CI: 1.087-1.793) effect against the development of TB.Conclusions:This study indicated that MBL2 polymorphisms, haplotypes, and diplotypes were associated with TB susceptibility in the Han Chinese population. Additionally, larger sample size studies are needed to further confirm these findings in the future.

  • 简介:Afterprimaryanalysesontheserumglycocon-jugatesoflungcancerandnormalindividulusingtheenzyme-linkedlectinassay(ELLA)with12kindsoflectins,PHAandLCAwereselectedtofurtherstudyintheseraof8kindsofcancers,4kindsofnon-malignantdiseaseand1kindofpostoperativecancer.Itwasfoundthatthetestvaluesof7kindsofcancerswithPHAorLCAweresignificantlyhigherthanthatofthenormal(P<0.01);thevaluesof4kindsofnon-malignantdiseaseswithPHAwerenothigher(P>0.05);thevaluesofthepostoperativecancerwithPHAwereobviouslylowerthanthatofthepreoperative(P<0.05).TheresultsshowedthattheserumglycoconjugateswhichcanbindtoPHAseemedrelatedtothecancerousexistenceinhumanbodies.Thesignificanceofthefindingswasdiscussed.

  • 简介:InsectshavealargefamilyofC-typelectinsinvolvedincelladhesion,pathogenrecognitionandactivationofimmuneresponses.Inthisstudy,32transcriptsencodingC-typelectindomainproteins(CTLDPs)wereidentifiedfromtheThitarodesxiaojinensistranscriptome.Accordingtotheirdomainstructures,sixCTLDPswithonecarbohydraterecognitiondomain(CRD)wereclassifiedintotheCTL-Ssubfamily.Theother23CTLDPswithtwoCRDsweregroupedintotheimmulectin(IML)subfamily.TheremainingthreewithextraregulatorydomainsweresortedintotheCTL-Xsubfamily.PhylogeneticanalysisshowedthatCTL-SandCTL-Xmembersfromdifferentinsectscouldformorthologousgroups.Incontrast,noT.xiaojinensisIMLorthologueswerefoundinotherinsects.Remarkablelineage-specificexpansioninthissubfamilywasobservedreflectingthattheseCTLDPs,asimportantreceptors,haveevolveddiversifiedmembersinresponsetoavarietyofmicrobes.PredictionofbindingligandsrevealedthatT.xiaojinensis,acoldadaptedspecies,conservedtheabilityofCRDstocombinewithCa^2+tokeepitsreceptorsfromfreezing.ComparativeanalysisofinductionofCTLDPgenesafterdifferentimmunechallengesindicatedthatIMLsmightplaycriticalrolesinimmunedefenses.ThisstudyexaminedT.xiaojinensisCTLDPsandprovidesabasisforfurtherstudiesoftheircharacteristics.

  • 简介:AbstractChronic obstructive pulmonary disease (COPD) has become the third-leading cause of death worldwide, which is a severe economic burden to the healthcare system. Chronic bronchitis is the most common condition that contributes to COPD, both locally and systemically. Neutrophilic inflammation predominates in the COPD airway wall and lumen. Logically, repression of neutrophilia is an essential fashion to COPD treatment. However, currently available anti-neutrophilic therapies provide little benefit in COPD patients and may have serious side effects. Thus, there is an urgent need to explore an effective and safe anti-neutrophilic approach that might delay progression of the disease. Sialic acid-binding immunoglobulin-like lectin (Siglec)-9 is a member of the Siglec cell surface immunoglobulin family. It is noteworthy that Siglec-9 is highly expressed on human neutrophils and monocytes. Ligation of Siglec-9 by chemical compounds or synthetic ligands induced apoptosis and autophagic-like cell death in human neutrophils. Furthermore, administration of antibody to Siglec-E, mouse functional ortholog of Siglec-9, restrained recruitment and activation of neutrophils in mouse models of airway inflammation in vivo. Given the critical role that neutrophils play in chronic bronchitis and emphysema, targeting Siglec-9 could be beneficial for the treatment of COPD, asthma, fibrosis, and related chronic inflammatory lung diseases.

  • 简介:BackgroundIt'sestablishedthatLectin-likeoxidizedlow-densitylipoproteinreceptor-1(LOX-1)isinvolvedinintimalhyperplasiaafterballooninjury.Therecentevidencealsosuggeststhatvalsartanhasanantiatheroscleticeffect.Inthisstudy,theexpressionofLOX-1andtheeffectofvalsartanonitsexpressionwasinvestigatedinrataortaafterballooninjury.MethodsRatmodelofaorticendothelialdenudationwasinducedby2Fballooncatheter.Ratswererandomlydividedintothreegroups:control,operationandvalsartantreatment.Theaortictissuesweretakenfromratsineachgroupondays14and28aftersurgery.ThethicknessofvascularwallwasmeasuredwithHEstain,LOX-1mRNAandproteinweredeterminedbyreversetranscription-polymersechainreaction(RT-PCR)andimmunohistochemistry,respectively.ResultsComparedwiththecontrolgroup,significantintimalthickeningwasobservedatday14and28afterinjury.Comparedwiththeoperationgroup,intimalthicknessofeachtimepointwassignificantlydecreasedinvalsartantreatmentgroup.Atday14and28afterballooninjury,theexpressionlevelsofLOX-1mRNAandproteinweresignificantlyincreased,andweregreatlydecreasedaftervalsartantreatment.ConclusionsTheexpressionofLOX-1isincreasedafterendothelialinjury.Valsartaninhibitsaorticintimalthickeninginducedbyendothelialdenudation,whichisassociatedwiththedownregulationofLOX-1expression.

  • 简介:摘要目的建立检测唾液酸化胎球蛋白A(Sia-fetuin A)的凝集素-酶联免疫吸附试验方法(lectin-ELISA),探讨其在原发性肝细胞癌(HCC)中的诊断价值。方法收集2017至2020年于上海东方肝胆外科医院就诊的HCC患者300例,疾病对照160例(包括肝硬化亚组36例、慢性乙型肝炎亚组124例),另同期收集健康体检者100名作为健康对照。根据接骨木凝集素(SNA)可与糖链末端α-2,6-半乳糖的唾液酸结合原理构建lectin-ELISA方法,检测各组血清Sia-fetuin A水平,以450 nm处吸光度值表示。采用logistic回归方法建立多指标联合检测模型,采用受试者工作特征曲线(ROC)评估单项指标及联合检测模型诊断HCC效能。结果建立了检测血清Sia-fetuin A的lectin-ELISA实验方法,其线性回归系数为0.978 5,精密度评估及干扰实验均符合临床检测要求。采用该方法检测各组血清Sia-fetuin A水平,HCC组、疾病对照组和健康对照组分别为1.362±0.310、1.199±0.370、1.086±0.420,3组之间依次降低。Sia-fetuin A、AFP和二者联合检测模型鉴别诊断HCC的曲线下面积分别为0.790、0.809和0.860,诊断模型敏感度79.3%(238/300),特异度95.0%(247/260)。HCC组300例患者中138例(46%)患者血清AFP为阴性(<20 μg/L),其血清Sia-fetuin A水平为1.364±0.305。将疾病对照组与健康对照组合并为非患癌组,其血清Sia-fetuin A水平为1.146±0.381。AFP阴性HCC患者血清Sia-fetuin A水平高于非患癌患者(t=6.134,P<0.001)。Sia-fetuin A与联合诊断模型诊断AFP阴性HCC的曲线下面积分别为0.776、0.919,联合诊断模型敏感度93.4%(129/138),特异度77.3%(201/260)。结论血清Sia-fetuin A及其联合检测模型可为AFP阴性HCC患者提供新的辅助诊断指标。
